The calibration of three Sartocheck filter testers |
The integrity test is the End User's method to confirm the structural integrity of a membrane filter before and after filtration. It is a non-destructive test that has been correlated to bacteria retention and validates the performance of the filter. According to the GMP guidelines and the requirements by the American health authority (FDA), manufacturers are obligated to test the filter for integrity before and after the production cycle. By performing a filter integrity test it's proved that the filter is fully functional and that no unwanted components did pass. To ensure a sterile product OBP uses two techniques namely a) filter sterilization and b) autoclaving. For all media preparations that are heat sensitive which contains proteins, amino acids and serum, sterilization of the product is obtained by filter sterilization as it cannot be autoclaved. Furthermore, air filters are used on all tanks and fermenters to ensure and confirm structural integrity and the sterility of products inside. To confirm that the filter membranes are still intact and that the correct pore sized membranes is used each filter is tested before and after each filtration process. The Sartocheck is the most important and only test system that is used daily to verify filter integrity. The filter integrity testing is the only accepted test method for the GMP and ISO accreditation for it also verify the filtered products integrity as well. Because of the internal and external pressure sensors that are very sensitive the Sartocheck should be calibrated quarterly. |

To appoint a service provider for a period of 12 months for Deep Cleaning Services (as and when required) |
To appoint a service provider for deep cleaning services at OBP for a period of not more than one year. This SOP is designed to provide guidance for environmental surface cleaning and disinfection of laboratories and work areas at OBP. Regular disinfection is required in any laboratory handling biohazardous agents. Regular disinfection is also important in other laboratories to reduce the risk of contamination. Reducing risk of contact exposure to infectious agents through routine cleaning and disinfection is an important part of regular laboratory environment maintenance. • Cleaning refers to the removal of dirt and impurities, including germs, from surfaces. Cleaning alone does not kill germs. But by removing the germs, it decreases their number and therefore any risk of spreading infection. Cleaning is achieved with soap and water. • Disinfecting works by using chemicals, for example EPA-registered disinfectants, to kill germs on surfaces. This process does not necessarily clean dirty surfaces or remove germs. But killing germs remaining on a surface after cleaning further reduces any risk of spreading infection. Below is the scope of work the service provider will be required to perform but not limited to points below • Prepare the workplace to facilitate cleaning and disinfection. For example, use plastic keyboard covers for shared computers, and disposable plastic clings over touch screens. • Visibly soiled surfaces must be cleaned prior to disinfection. • Disinfectants have a finite shelf life. Do not use expired disinfectants. • Disinfectants present health and physical hazards. Read and adhere to all precautions and instructions (including proper use concentration) in the manufacturer’s Safety Data Sheet (SDS) and label. • Disinfectant efficacy depends on proper contact time to achieve adequate kill time. This varies between disinfectants. Ensure proper contact time during use. • Pause lab operations in the target area during cleaning/disinfection. • Use damp cleaning methods. Do not clean with dry dusting or sweeping as this may create airborne particles. • Clean from least to most dirty areas. Change mop heads, rags, and similar items frequently during the cleaning/disinfection process. Consider using disposable cleaning items. • Use a double-bucket method when rinsing is necessary (one bucket for cleaning/disinfectant solution, one for rinsing). Some disinfectants are corrosive or otherwise damaging to certain surfaces and will require rinsing after the appropriate contact time has been achieved. • Clean, disinfect, and dry non-disposable equipment at the end of each work shift and when known to be contaminated. When risk of community acquired infection is high, clean and disinfect shared spaces/items after use and before sharing with others. Work surfaces (e.g., biosafety cabinets, lab benches, etc.) and high-touch surfaces should be cleaned and disinfected. Examples include: o Door, appliance, and cabinet handles; o Light switches; o Benchtops; o Desks; o Chairs/seats; o Telephones; o Computers, screens, keyboards, and other analytical equipment (e.g., centrifuge lids, biosafety cabinets, fume hoods, incubators, etc.); o Reagent and other containers, pipettors, and other shared equipment/devices o Hand-washing sinks and handles. DISINFECTION Use all disinfectants according to manufacturer’s instructions. Proper disinfection procedure should include: • Recommended dilution of disinfectant; • Liberal application with clean cloth, sponge, or hand mop; • Sufficient surface contact time as indicated by manufacturer; • Allow to air dry. |

To obtain approval for the procurement of 30 x 0.1 micron capsule final filter |
All viral and bacterial vaccines are being filtered through a 0.45/0.2 µn pre and 0.45/0.2 µn and 0.2/0.1-micron final filter. For oils and serum, we start filtering with a 0.3-micron pre filter. Up to 45 batches per week are filtered depending on the production schedule. The filters for the Water Plant and Softener vary from 0.2 µn 5 inch up to 20 inch and the water filters from 8 µn to 90 µn filters. There is a 0.2 µn air filter on top of each 50L tank. The water filters protect the RO membranes, and the air filters are protecting the tanks from pressure build up and collapsing. The air filter protects the filtration tank from collapsing after sterilization and helps when media is filtered into the tank. All filters used by Raw Material needs to comply to the following standards: 1- Products must conform and meet all FDA requirements including CFR21. 2- Products must meet all Biological Safety requirements for USP Class 6 for plastics at 121Deg C 3- Products must be accompanied with full Validation guide which must have full description of the following section USP Class VI for plastic MEM Elution Extractables substances (What filter can add to product being filtered) Full correlation of integrity testing with liquid bacterial challenge Log reduction values should be >7/cm2 of filter area. (This is amount of bacteria /cm2) 4- Filters supplied must come with complete COC including integrity test data (For Gas filters WIT data) 5- For liquid filters membranes need to provide high flow and low protein binding as membranes with high binding capacities may remove actives from product being filtered. Preferable membranes are Cellulose Acetate and Polyethersuplhone. All filters should be pre-flushed with ultra-pure water before final assembly. Products at OBP all have high protein actives and therefore it is essential that membranes used have low binding capacity. For air filters membranes need to be PTFE to prevent membranes from wetting out and there preventing gas flow. 6-Integral prefilter layer is a big advantage as it saves the cost of a pre-filter. 7- Cartridges should show data regarding regenerations for re-use and a minimum autoclave cycle at 121Deg C of 30 cycles. The following mediums are using the following filters: Bacterial medium Final Filter Pre-Filter TAS 0.2/0.45 µn 0.3 µn Leukopast 0.2/0.45 µn 0.2/0.45 µn RPMI medium 0.2/0.45 µn 0.3 µn Oils 0.2/0.45 µn Bot C and D medium 2 x 0.3 µn Saline 0.2/0.45 µn Saponin 0.2/0.45 µn 0.2/0.45 µn Viral medium Eagles 0.2/0.1 µn 0.3 or 0.2/0.45 µn Peptone Sucrose 0.2/0.45 µn 0.2/0.45 µn BLP 0.2/0.45 µn 0.2/0.45 µn 2SP 0.2/0.45 µn 0.2/0.45 µn ATV 0.2/0.1 µn 0.3 or 0.2/0.45 µn Alsevers 0.2/0.1 µn 0.3 or 0.2/0.45 µn Dispase 0.2/0.1 µn 0.3 or 0.2/0.45 µn Earls 0.2/0.1 µn 0.3 or 0.2/0.45 µn Penicillin 0.2/0.45 µn PBS Dulbecco 0.2/0.45 µn PBS Blood 0.2/0.45 µn 0.2/0.45 µn PBS without Ca / Mg 0.2/0.45 µn 0.2/0.45 µn Sterile Water 0.2/0.45 µn Streptomycin 0.2/0.45 µn Gentamycin 0.2/0.45 µn Phenol Saline 0.2/0.45 µn 0.2/0.45 µn Water plant and Softener Ro Plant 8 µn Water filter 0.2 µn air filter 10inch WFI plant 0.2 µn air filter 5 inches 0.2 µn air filter 20 inches Water Softener 90 µn Water filter 20 inches 50L Tank air filters 50 L Filtration tanks 0.2µn |
