These kits usually work on the principle of enzyme-linked immunosorbent assay or a similar immunoassay format. They detect the protease-resistant prion protein in tissue samples brainstem/obex tissue. Normal prion proteins are degraded by proteinase K, whereas PrP^Sc is resistant and remains detectable by monoclonal antibodies in the kit.
